  • A new exercise on lysozyme employs egg white as a source of this enzyme to demonstrate that lysozyme breaks down peptidoglycan in bacterial cells, making the bacteria prone to lysis. This exercise will demonstrate the difference in sensitivity of Gram-positive and Gram-negative bacteria to lysozyme as a result of the presence of the outer membrane that surrounds Gram-negative cells.   • The exercises on the identification of an unknown bacterium have been rewritten and reorganized into exercises that separate the tests into categories based on fermentation, oxidation, degradative and hydrolytic enzymes, and multiple test media.
